The crystallization temperature used in the production of polycrystalline films is inadequate for the formation of epitaxial films. Ultra-processed foods, abundant in fat and sugar, are a major contributing factor to the worldwide prevalence of the Western diet. This preference stems from their often lower cost and ease of consumption compared to preparing fresh, nutritious food. Consumption of UPF has been linked by epidemiological research to obesity, non-alcoholic fatty liver disease (NAFLD), and insulin resistance. In molecular research, mice on Western diets have been used to characterize signaling pathways in these diet-induced conditions. Nevertheless, these investigations subjected mice to constant dietary regimens, a practice inconsistent with the sporadic consumption patterns observed in natural environments. We compared the effects of a high-fat, high-sucrose diet administered weekly in mice to those fed a continuous high-fat, high-sucrose diet or a standard diet. In comparison to the control group, our results show oral glucose tolerance tests (oGTTs) in animals were impaired after consuming a high-fat, high-sugar (HFHS) diet for a single day. The impairment was reversed after 24 hours of consuming a regular diet, but weekly high-fat, high-sugar consumption worsened the condition. More specifically, the oral glucose tolerance test (oGTT) impairment remained after 12 weeks, even with 6 days on a controlled diet. Observational studies of animal groups consuming a high-fat, high-sugar diet (HFHS) weekly versus continuously revealed comparable outcomes in regards to liver steatosis, inflammation, impaired insulin signaling, and endoplasmic reticulum stress, but the weekly fed animals displayed diminished weight gain. The robustness and statistical significance of the water efflux rate constant (k(io)), determined from a two-flip-angle Dynamic Contrast-Enhanced (DCE) MRI protocol, are assessed in a murine glioblastoma model at 7 Tesla in this manuscript. The repeatability of kio measurements and contrast kinetic parameters was examined through a test-retest experiment with seven participants. The cellular metabolic associations of kio were investigated using DCE-MRI and FDG-PET techniques in a cohort of 7 subjects. Contrast kinetic parameters and kio (n=10) were used to monitor tumor response to the combination therapy of bevacizumab and fluorouracil (5FU). In test-retest experiments, compartmental volume fractions (ve and vp) were found to be consistent across different scan instances; however, vascular functional measurements (Fp and PS) and kio displayed considerable changes, potentially attributable to physiological transformations of the tumor. Regarding tumors, their standardized uptake values (SUV) demonstrate a linear correlation with kio (R² = 0.547), a positive correlation with Fp (R² = 0.504), and weak correlations with ve (R² = 0.150), vp (R² = 0.077), PS (R² = 0.117), Ktrans (R² = 0.088) and whole tumor volume (R² = 0.174). The kio in the treated group was substantially lower than the control group's value post-bevacizumab, measurable one day after treatment.
